What is Amlodipine
Amlodipine is a long-acting 1,4-dihydropyridine calcium channel blocker. It acts primarily on vascular smooth muscle cells by stabilizing voltage-gated L-type calcium channels in their inactive conformation. By inhibiting the influx of calcium in smooth muscle cells, amlodipine prevents calcium-dependent myocyte contraction and vasoconstriction. A second proposed mechanism for the drugs vasodilatory effects involves pH-dependent inhibition of calcium influx via inhibition of smooth muscle carbonic anhydrase. Some studies have shown that amlodipine also exerts inhibitory effects on voltage-gated N-type calcium channels. N-type calcium channels located in the central nervous system may be involved in nociceptive signaling and pain sensation. Amlodipine is used to treat hypertension and chronic stable angina.
Amlodipine mechanism of action
Amlodipine decreases arterial smooth muscle contractility and subsequent vasoconstriction by inhibiting the influx of calcium ions through L-type calcium channels. Calcium ions entering the cell through these channels bind to calmodulin. Calcium-bound calmodulin then binds to and activates myosin light chain kinase (MLCK). Activated MLCK catalyzes the phosphorylation of the regulatory light chain subunit of myosin, a key step in muscle contraction. Signal amplification is achieved by calcium-induced calcium release from the sarcoplasmic reticulum through ryanodine receptors. Inhibition of the initial influx of calcium decreases the contractile activity of arterial smooth muscle cells and results in vasodilation. The vasodilatory effects of amlodipine result in an overall decrease in blood pressure. Amlodipine is a long-acting CCB that may be used to treat mild to moderate essential hypertension and exertion-related angina (chronic stable angina). Another possible mechanism is that amlodipine inhibits vascular smooth muscle carbonic anhydrase I activity causing cellular pH increases which may be involved in regulating intracelluar calcium influx through calcium channels.

What is Norvasc?
Norvasc is a calcium channel blocker that dilates blood vessels and improves circulation. Norvasc is used to treat chest pain (angina) and other conditions caused by coronary artery disease. Norvasc is also used to treat high blood pressure (hypertension). Lowering blood pressure can reduce the risk of stroke or heart attack. Amlodipine is intended for adults and children who are at least 6 years old.
Before taking Norvasc, tell your doctor if you have congestive heart failure or liver disease. Drinking alcohol can lower blood pressure even more and may increase some side effects of Norvasc. If you are being treated for high blood pressure, continue using Norvasc, even if you feel well. High blood pressure often has no symptoms. You may need to use blood pressure medication for the rest of your life.
Norvasc is only part of a complete treatment program, which may also include diet, exercise, weight control, and other medications. Watch your diet, medications and exercises very carefully. Tell your doctor about all other heart or blood pressure medications you are taking. Chest pain may worsen when you first take this medicine or when your dose is increased. Call your doctor if chest pain is severe or continues.
What is Tribenzor?
Tribenzor contains a combination of amlodipine, hydrochlorothiazide, and olmesartanan. Amlodipine is a calcium channel blocker that relaxes (dilates) blood vessels and improves circulation. Hydrochlorothiazide is a thiazide diuretic (water tablet) that helps prevent your body from absorbing too much salt, which can cause fluid retention.
Olmesartan is an angiotensin II receptor antagonist that keeps blood vessels from narrowing, which lowers blood pressure and improves blood flow. Tribenzor is used to treat high blood pressure (hypertension). Tribenzor is usually given after other blood pressure medications have been tried without successful treatment.
You should not take Tribenzor if you are allergic to sulfa drugs, or if you cannot urinate. Do not use Tribenzor if you are pregnant. If you become pregnant, stop taking this medicine and tell your doctor right away. If you have diabetes, do not use Tribenzor with any medicine that contains aliskiren (Amturnide, Tekturna, Tekamlo).
