Orally administered cholic acid is absorbed by passive diffusion through plasma membranes along the length of the gastrointestinal tract, because of its hydrophobicity. Glycol- and taurine-conjugated bile acids are absorbed at ileum through receptormediated transportation (apical sodium-dependent bile-acid transporter, ASBT). In the presence of enteral bacteria, a portion of cholic acid may be dehydroxylated to form secondary bile acid. Absorbed cholic acid enters blood stream of portal vein, and is taken up by hepatocyte via Na+ -dependent and Na+ -independent transporters.

What is Cholic Acid
A major primary bile acid produced in the liver and usually conjugated with glycine or taurine. It facilitates fat absorption and cholesterol excretion.Cholic acid, formulated as Cholbam capsules, is approved by the United States Food and Drug Administration as a treatment for children and adults with bile acid synthesis disorders due to single enzyme defects, and for peroxisomal disorders (such as Zellweger syndrome).

Indication of Cholic Acid
Oral cholic acid is indicated for: treatment of bile acid synthesis disorders due to single enzyme defects; and as adjunctive treatment of peroxisomal disorders including Zellweger spectrum disorders in patients who exhibit manifestations of liver disease, steatorrhea or complications from decreased fat soluble vitamin absorption.
Toxicity of Cholic Acid
Cholic acid is a primary bile acid. Primary bile acids are biosynthesized in the liver and are key constituents of normal bile. The dose of cholic acid to be administered to the patients is intended to restore a concentration that is equivalent to that physiologically present in humans.
