Ethinyl Estradiol volume of distribution
After a wash-out period of one week, the same preparation was administered during a treatment period of three months. After single dose administration, maximum concentrations of CPA in the serum were 15.2 +/- 6.6 ng/ml. Post maximum drug levels declined biphasically with half-lives of 0.8 +/- 0.4 h and 54.0 +/- 26.0 h, respectively. The apparent clearance was calculated to be 3.6 +/- 0.9 ml x min-1 x kg-1 and the volume of distribution (Vz) was 986 +/- 4371. The free fraction of CPA was 3.5 +/- 1.9% and the fractions bound to heat labile proteins and albumin were 4.6 +/- 2.2% and 92.0 +/- 3.5%, respectively. Trough levels of CPA in the serum increased during a treatment cycle, reaching a steady-state around day 16. An about two-fold accumulation of CPA was observed, which was less than expected theoretically. SHBG concentrations in the serum increased by a factor of three during a cycle, without having any effect on the protein binding of CPA. At the end of treatment cycle three, the terminal half-life of CPA had increased to a mean value of 78.6 +/- 16.0 h and the volume of distribution to a value of 1304 +/- 427 1. The apparent clearance showed a small, although significant decrease to a value of 3.0 +/- 0.4 ml x min-1 x kg-1. The observed changes Vz and t 1/2 during the treatment period were attributed to the distribution of CPA into a deep compartment and the slow release of the drug from this compartment.

What is Ethinyl Estradiol
A semisynthetic alkylated estradiol with a 17-alpha-ethinyl substitution. It has high estrogenic potency when administered orally and is often used as the estrogenic component in oral contraceptives. Ethinyl estradiol is marketed mostly as a combination oral contraceptive under several brand names such as Alesse, Tri-Cyclen, Triphasil, and Yasmin. The FDA label includes a black box warning that states that combination oral contraceptives should not be used in women over 35 years old who smoke due to the increased risk of serious cardiovascular side effects.
Ethinyl Estradiol mechanism of action
Estrogens diffuse into their target cells and interact with a protein receptor. Target cells include the female reproductive tract, the mammary gland, the hypothalamus, and the pituitary. Estrogens increase the hepatic synthesis of sex hormone binding globulin (SHBG), thyroid-binding globulin (TBG), and other serum proteins and suppress follicle-stimulating hormone (FSH) from the anterior pituitary. This cascade is initiated by initially binding to the estrogen receptors. The combination of an estrogen with a progestin suppresses the hypothalamic-pituitary system, decreasing the secretion of gonadotropin-releasing hormone (GnRH).

Indication of Ethinyl Estradiol
For treatment of moderate to severe vasomotor symptoms associated with the menopause, female hypogonadism, prostatic carcinoma-palliative therapy of advanced disease, breast cancer, as an oral contraceptive, and as emergency contraceptive.
Toxicity of Ethinyl Estradiol

What is a Lo Loestrin Fe?
Lo Loestrin Fe (ethinyl estradiol, norethindrone and black fumarate) is a combination drug containing female hormones that prevent ovulation (the release of an egg from the ovary). This drug also causes changes in your cervical mucosa and uterus, which complicates the penetration of sperm into the uterus and more difficult for a fertilized egg that attaches to the uterus. Lo Loestrin Fe is used as a contraceptive to prevent pregnancy.
Do not use Lo Loestrin Fe if you are pregnant or have recently had a baby. You should not take Lo Loestrin Fe if you have any of the following conditions: uncontrolled high blood pressure, heart disease, blood clotting problems, circulatory problems, diabetic problems with the eyes or kidneys, unusual vaginal bleeding, liver disease or cancer liver, severe migraine headaches, if you smoke and are over 35 years old, or if you have ever had breast or uterus cancer, jaundice caused by birth control pills, heart attack, stroke or blood clot.
You may have to use backup birth control, such as condoms or spermicide, when you first start using Lo Loestrin Fe or skip a dose. Follow your doctor’s instructions. Lack of pills increases the risk of becoming pregnant. Carefully follow the “missed dose” instructions if you forget to take your Lo Loestrin Fe tablets.
Some drugs can make Lo Loestrin Fe less effective in preventing pregnancy, including antibiotics, drugs for hepatitis C, drugs for HIV/AIDS, drugs for seizures, or barbiturate sedatives. Tell your doctor about all other medicines that you use.
