Interferon beta-1a volume of distribution
Systemic clearance values in monkeys, rats, and mice decreased, respectively, from 232, 261, and 247 ml/h/kg for the unmodified IFN-beta-1a to 30.5, 19.2, and 18.7 ml/h/kg for the PEGylated form, while volume of distribution values decreased from 427, 280, and 328 ml/kg to 284, 173, and 150 ml/kg. The decreased clearance and volume of distribution resulted in higher serum antiviral activity in the PEG IFN-beta-1a-treated animals. In the rat, a more extensive set of dosing routes was investigated, including intraperitoneal, intratracheal, and oral administration.

What is Interferon beta-1a
Human interferon beta (166 residues), glycosylated, MW=22.5kD. It is produced by mammalian cells (Chinese Hamster Ovary cells) into which the human interferon beta gene has been introduced. The amino acid sequence is identical to that of natural human interferon beta.
Interferon beta-1a mechanism of action
Interferon beta binds to type I interferon receptors (IFNAR1 and IFNAR2c) which, upon dimerization, activate two Jak (Janus kinase) tyrosine kinases (Jak1 and Tyk2). These transphosphorylate themselves and phosphorylate the receptors. The phosphorylated INFAR receptors then bind to Stat1 and Stat2 (signal transducers and activators of transcription) which dimerize and activate multiple (~100) immunomodulatory and antiviral proteins. Interferon beta binds more stably to type I interferon receptors than interferon alpha.

Toxicity of Interferon beta-1a
The manufacturer of interferon beta-1a (Rebif) states that, although patients receiving interferon beta-1a may receive concomitant vaccination, the overall effectiveness of vaccination in patients receiving interferon beta has not been determined. In a nonrandomized study in patients with multiple sclerosis, those receiving interferon beta-1a (Rebif; 12 million units (44 ug) 3 times weekly) for at least 6 months were vaccinated with influenza virus vaccine inactivated and the seroconversion rate was compared to that in patients not receiving the drug.
