What is Naloxegol
Naloxegol, for “PEGylated naloxol” is a peripherally-selective opioid antagonist developed by AstraZeneca. It was approved by the FDA in September 2014 and is indicated for the treatment of opioid-induced constipation (OIC) in adult patients with chronic non cancer pain. The advantage of naloxegol over the opioid antagonist naloxone is that its PEGylated structure allows for high selectivity for peripheral opioid receptors and lack of entry into the central nervous system through the blood-brain barrier.
Naloxegol mechanism of action
Naloxegol is an antagonist of opioid binding at the mu-opioid receptor. When administered at the recommended dose levels, naloxegol functions as a peripherally-acting mu-opioid receptor antagonist in tissues such as the gastrointestinal tract, thereby decreasing the constipating effects of opioids. Naloxegol has shown more than 6000 fold selectivity for the peripheral mu receptors, and its PEGylated form restricts its action only to the periphery, not affecting the pain-relieving mechanism of opioids in the central nervous system.

What is Movantik?
Movantik is a prescription drug that blocks certain effects of narcotic drugs. An opioid is sometimes called a drug. Movantik is used to treat opioid induced constipation (AEI) caused by prescribed drugs, called drugs or opiates, in adults with prolonged (chronic) pain that is not caused by cancer. Naloxegol treats constipation without diminishing the analgesic effects of the drug.
Movantik is used in people who have taken a narcotic painkiller for at least 4 weeks to treat chronic pain that is not caused by cancer.
You should not use Movantik if you have a blockage in your stomach or intestines. Tell your doctor if you have opioid withdrawal symptoms, such as anxiety, irritability, sweating, chills, yawning, stomach pain, and diarrhea. Stop using this medicine and consult a doctor immediately if you have severe stomach pain and / or diarrhea (which can lead to serious medical problems).
Many drugs can interact with naloxol, and some drugs should not be used together. Tell your doctor about all the medicines you use and those you start or stop using during your treatment with Movantik.
